Catalytic intermediates of cytochrome bd terminal oxidase at steady-state: ferryl and oxy-ferrous species dominate.

The cytochrome bd ubiquinol oxidase from Escherichia coli couples the exergonic two-electron oxidation of ubiquinol and four-electron reduction of O(2) to 2H(2)O to proton motive force generation by transmembrane charge separation. The oxidase contains two b-type hemes (b(558) and b(595)) and one heme d, where O(2) is captured and converted to water through sequential formation of a few interme...

The roles of the two proton input channels in cytochrome c oxidase from Rhodobacter sphaeroides probed by the effects of site-directed mutations on time-resolved electrogenic intraprotein proton transfer.

The crystal structures of cytochrome c oxidase from both bovine and Paracoccus denitrificans reveal two putative proton input channels that connect the heme-copper center, where dioxygen is reduced, to the internal aqueous phase. In this work we have examined the role of these two channels, looking at the effects of site-directed mutations of residues observed in each of the channels of the cyt...

Characterization of a novel g' = 2.95 EPR signal from the binuclear center of mitochondrial cytochrome c oxidase.

The oxidized binuclear heme a3/CuB center of slow forms of bovine cytochrome oxidase exhibits a characteristic EPR signal at g' = 12. Following the (rapid) dithionite reduction of heme a and CuA, an additional EPR signal becomes apparent at g' = 2.95. As electrons enter the binuclear center this signal decays at the same slow rate as the g' = 12 signal. In the fully oxidized slow enzyme the sma...

The structure of a ferrous heme-nitro species in the binuclear heme a3/CuB center of ba3-cytochrome c oxidase as determined by resonance Raman spectroscopy.

Members of the cytochrome c oxidase family exhibit nitrite reductase activity. In this work, we have characterized a ferrous heme a3-nitro species in ba3-oxidase by resonance Raman spectroscopy. This provides the first evidence for the structure of a nitrite-bound species in the binuclear heme/copper center of cytochrome c oxidases.

Rational engineering of Thermus thermophilus cytochrome c552 to a thermally tolerant artificial peroxidase.

In a previous study, we constructed a prototype thermally tolerant artificial peroxidase from an electron transfer protein, cytochrome c(552) from Thermus thermophilus, and demonstrated that engineering of proteins from thermophiles could be a promising methodology to produce artificial enzymes for practical use. In the present study, further improvement of the prototype (the V49D/M69A mutant) ...
